En 1961, el médico australiano WG McBride escribió una carta al director en The Lancet titulada Talidomida y anomalías congénitas. En ella advertía de la posible relación entre varios casos de malformaciones en bebés y el consumo de esta sustancia, muy común para tratar las náuseas y vómitos matutinos durante los primeros meses del embarazo. Hoy sabemos que más de 10.000 niños de todo el mundo nacieron con serias deformaciones en brazos y piernas a causa de este medicamento.
La tragedia de la talidomida cambió de manera rotunda el modo en que los fármacos se desarrollan, prueban y regulan. En Estados Unidos, por ejemplo, se amplió la autoridad de la Food and Drug Administration y en muchos países hubo un importante endurecimiento de las pruebas que debían pasar los medicamentos antes de ser comercializados.
Esta carta que McBride envió a The Lancet es el primero de los 27 artículos que contiene esta publicación de la Fundación Dr. Antonio Esteve y que forman parte de la columna vertebral de la Farmacoepidemiología. Esta disciplina se encarga de estudiar los efectos beneficiosos o perjudiciales y del uso de medicamentos en poblaciones humanas, utilizando para ello los métodos y el razonamiento epidemiológicos. Como ámbito de estudio definido, nació a mediados de los ochenta, consolidándose en 1989 con la creación de la Sociedad Internacional de Farmacoepidemiología. Esta institución agrupa a especialistas tanto de universidades y hospitales como de agencias del medicamento, autoridades sanitarias y compañías de la industria biomédica.
De esta sociedad internacional fue presidenta Susana Pérez Gutthann, la encargada de seleccionar, junto a algunos de sus colegas epidemiólogos, los artículos que, a su juicio, han dado forma a la Farmacoepidemiología. Este volumen número seis de la serie Pharmacotherapy Revisited recoge, pues, en edición facsímil, tal como se publicaron en su momento, algunos de los artículos más representativos de esta disciplina.
